What is an explant?

By the end of this lesson

  • Define an explant
  • Distinguish tissues used to start cultures
  • Record the source and state of selected tissue

See it

An explant is the plant material selected to begin a culture. Depending on the plant and method, an explant may be a node, shoot tip, meristematic region, leaf piece, root piece, embryo, or other tissue. The exact source matters because different tissues can carry different contamination risks and regenerative capacities.

Understand it

Define an explant

Record species/cultivar and the exact tissue used, not only the common plant name.

An explant is the starting plant tissue or organ, such as a node, shoot tip or leaf piece.

Distinguish tissues used to start cultures

Explant age, health, source environment, and developmental state can affect response.

Tissue identity matters because regenerative capacity and contamination context can differ between explant types.

Record the source and state of selected tissue

A protocol for one explant type should not silently be applied to another.

Identify the plant and exact starting tissue so a later comparison does not confuse different explant strategies.

Worked example

A blackberry node and a leaf piece come from one donor plant. They are different explants, even though the common plant name is the same. A protocol tested on the node does not automatically describe what will happen to the leaf piece.

Explain your decision before opening the worked reasoning below.

Show the worked reasoning

A change in explant is a change in biological context and must be explicit rather than silently inheriting evidence.

The physiological condition and environment of source material can affect contamination risk and culture response.

Important boundary

“Blackberry” is not enough protocol context; cultivar, explant, and stage can matter.

Try it

What should accompany the node in the example's record?

Use the worked example above to explain your answer in your own words before taking the quiz.

Lesson quiz

Assessment version: 2026-10-15-v2. Previous reading-completion markers are retained. This revised quiz has its own result.

Apply it

Use the worked example to write a short decision record: what did you observe, which rule did you apply, and what would you check before acting?

Completion marks reading progress, not a quiz pass or professional certification.
